How can I make the distinction between "don't know" prononciation "dunno" and expression " do know" ?

Hello,
When you say : I do know that you are guilty, you insist and so the do will be heavily stressed [du:]
When you say "I dunno", the first syllable will be very short : [dn]
So there is no mistaking one for the other if you pronounce them right.
